Pipetting Techniques

Overview

  • Two basic pipetting techniques:
    1. Forward Pipetting
    2. Reverse Pipetting

Forward Pipetting Technique

  • Common technique used in many laboratories.

Reverse Pipetting Technique

  • Description:
    • The operation button is pressed to the second stop before aspirating liquid.
    • This action aspirates an excess (access) volume of liquid.
  • Dispensing Process:
    • The operation button is pressed only to the first stop to dispense the target volume.
    • The access volume is either returned to the source or discarded.

Benefits of Reverse Pipetting

  • Improved Accuracy:
    • Particularly beneficial for dispensing viscous, volatile, and foaming liquids.
  • Enhanced Repeatability:
    • Small volumes are dispensed more accurately.
  • Reduces Errors:
    • Liquid sticking to the tip or evaporating during the operation does not affect the target volume.

Conclusion

  • Knowing and utilizing both pipetting techniques can enhance precision in laboratory settings.
