Investigating Temperature's Impact on Catalase

In this experiment we will be investigating the effect of temperature on the rate of catalase activity. For this experiment you are going to need an enzyme source. Here we have celery, but you may also use radishes. Hydrogen peroxide. pH 9 buffer solution. Washing up liquid. Boiling tubes. Graduated cylinders. Water Bats ranging from 0 to 60 degrees Celsius A Thermometer A Knife A Chopping Board An Electronic Balance Weigh Boats A Dropper And a Test Tube Rack Add 20cm3 of the PH Buffer 9 to the graduated cylinder Use a dropper for the final 1-2cm cube to ensure the bottom of the meniscus rests on the calibration mark. Add one drop of washing off liquid to the solution. Finely chop 5g of celery. Weigh the chopped celery on a weigh balance and as close as you can to 5 grams and record your actual result. Add 2cm3 of hydrogen peroxide to a boiling tube. Add 5 grams of the finely chopped celery to the cylinder. The graduated cylinder and the boiling contain the hydrogen peroxide in a nice trabate until zero degrees is reached. Add the hydrogen peroxide from the boiling tube into the graduated cylinder and record the initial reading immediately. Let's remove the graduated cylinder from the output and record the volume. Repeat the procedure for different temperatures ranging from 5 to 60 degrees Celsius. For example, 25 degrees Celsius, 45 degrees Celsius and 60 degrees Celsius. When you have all your results collected, you should fill it out in a table such as the one we have here. We have temperature in degrees Celsius. We have initial volume, centimetres cubed, centimetres cubed, and volume of foam produced, centimetres cubed. Once you have filled out your table of results, you can now put them into a graph. On the y-axis we have enzyme activity, which is the volume of foam produced after two minutes. On the x-axis we have temperature in degrees Celsius. The highest point in the graph is known as the optimum temperature, which is the temperature at which your enzyme works best.
