[Music] [Music] how to pour plates sterile plates and a hundred milliliter bottle of sterile tsa agar are provided the plates are left stacked the Egger should still be molten but the bottle should be at a comfortable temperature to hold in your hand open the bottle and flame the neck of the bottle here you can place the cap down with the open side up in the updraft of the Bunsen flame lifting the lid of the bottom plate pour approximately 20 to 25 milliliters of molten agar in to the plate replace the lid flame and the neck of the bottle now lift the lid of the second place and pour in 20 to 25 mils of molten agar and replace the lid this is repeated until all plates have been filled the agar must now be allowed to solidify which typically takes 30 minutes or longer you will notice the agar turns opaque as it cools and solidifies
