Compound Light Microscope: Parts and Operation

Key Components

  • Arm and Base

    • Use both to hold and support the microscope securely.
  • Oculars (Eyepieces)

    • Adjustable to fit the width of the eyes.
    • Magnification: 10x.
  • Revolving Nose Piece

    • Holds multiple objective lenses.
    • Different magnifications available: 10x, 40x, and 100x.
  • Stage

    • Holds the slide in place via a clamp.
    • Includes stage moving controls:
      • Moves stage back and forth, right and left.
  • Adjustment Dials

    • Coarse Adjustment:
      • Moves stage quickly for rough focusing.
    • Fine Adjustment:
      • Moves stage in small increments for fine focusing.
  • Condenser

    • Located under the stage.
    • Controls and focuses light on the specimen.
    • Best position is typically all the way up.
  • Iris Diaphragm Lever

    • Located on the condenser.
    • Adjusts the amount of light passing through.
  • Light Source

    • Located below the condenser.
    • Front switch to turn on/off.
    • Side dial to adjust brightness.
    • Start with low light intensity.

Operating the Microscope

  1. Initial Setup

    • Start with the lowest power objective.
    • Adjust light intensity to low initially.
  2. Focusing

    • Use coarse adjustment for initial focus.
    • Fine-tune with fine adjustment.

Cleaning and Storage

  • Cleaning

    • Use lens paper to clean oculars and objectives.
    • Lens cleaner may be necessary for removing oil.
  • Storage

    • Diminish light intensity and turn off the power.
    • Rotate nose piece to low or no power objective.
    • Lower the stage completely.
    • Wrap power cord around the base/arm.
    • Use dust cover if available.
