Absolute Configurations
Introduction
- Absolute configurations are designations given to chiral centers.
- This lecture is part of a broader chapter on isomers and stereochemistry.
- Key Terms:
- Chiral Center: Carbon atom with four different groups attached.
- Mirror Image Forms: Two forms of a chiral center that are mirror images.
- Cahn-Ingold-Prelog System: Set of rules to designate forms of chiral centers as R or S.
Cahn-Ingold-Prelog System
- Assigns priorities to the groups attached to a chiral center based on atomic numbers.
- Priority Assignment:
- Assign numbers 1 (highest) to 4 (lowest) based on atomic number.
- Ties in atomic numbers are resolved by looking at atoms further along the chain.
Determining R and S Configurations
- Right-Handed Turn (R): Corresponds to the Latin word 'Rectus'.
- Left-Handed Turn (S): Corresponds to the Latin word 'Sinister'.
- Steps:
- Start at priority 1 and go to 2, then to 3 (ignore 4).
- If the turn is right-handed and the lowest priority group (4) is in the back, it's R.
- If the turn is left-handed, it's S.
Special Cases
- Lowest Priority Not in Back:
- If it's a wedge bond (coming out), a right-handed turn means S (opposite perspective).
- If it's in the plane of paper: worst scenario for students; approach depends on perspective.
Naming Molecules with Chiral Centers
- Name as usual, then add chiral center configuration.
- Example: "2-chlorohexane" needs more detail if there are chiral centers.
- Designation included in parentheses at the beginning of the name, e.g., (R)-2-chlorohexane.
- Multiple chiral centers require location numbers, e.g., (2R, 3S)-2-chloro-3-methylhexane.
Examples
- Single Chiral Center:
- Assign priorities using atomic numbers and position.
- Determine R/S configuration based on turn direction.
- Multiple Chiral Centers:
- Assign and compare priorities for each center.
- Determine and denote configurations in compound names.
Conclusion
- Understanding absolute configurations is crucial in distinguishing between different stereoisomers.
- Practice is key to mastering the assignment of R and S.
